introduction | ethyl 3-(3-fluorophenyl)-3-carbonyl propionate is also called ethyl 3-fluoro-β-oxo-phenylpropionate (keto ester), which can be prepared by the reaction of 3-fluorobenzoic acid and ethyl malonate potassium. |
Preparation | A-CDI(2.6g) was added in batches to a stirred thick slurry of 3-fluorobenzoic acid (1.2g) in CH3CN(6mL). Stir the resulting turbid solution for 4 hours (raise the batch temperature to 22°C). Flask B- Fill 100mL round bottom flask with ethyl malonate potassium (3.17g) and CH3CN(30 mL). MgCl2(1.47g) was then added in batches for 15 minutes. The mixture was stirred at 35°C for 30 minutes, and then cooled to 25°C. Slowly add triethylamine (TEA)(6mL) and stir the slurry for 30 minutes. Then the solution in flask A was transferred to the slurry in flask B after 5 minutes. Stir the reaction mixture for 1.5 hours, cool to 7°C, and then quench with 3N HCl(32mL) (internal temperature maintained < 20°C). Concentrate the resulting two-phase solution and extract the resulting concentrate with MTBE. Collect organic phase and wash with water, 5% NaHCO3 and finally 20% NaCl. Then the organic layer was concentrated under reduced pressure to obtain ethyl 3-(3-fluorophenyl)-3-carbonylpropionate, an orange oil (2.921g) with a yield of 97%. |
